硫酸及硝酸溶液對化學性防護手套之滲透機制探究

背景:硫酸及硝酸普遍使用於電子零組件製造及化學原料製造等產業。根據勞動部職業安全衛生署職業傷病管理服務中心統計,接觸性皮膚炎占總通報病例數的8.2%,為排名第三之職業病。製造業勞工為手部接觸性皮膚炎的好發族群,可能與接觸酸性溶液有關。因此,選用及穿戴適合的化學性防護手套做為降低皮膚暴露的最後一道防線是重要的。目的:為評估化學防護手套對酸性溶液的防護能力,本研究參考ASTM F739-12進行滲透試驗,同步以酸鹼度計(pH meter)及離子交換層析儀(ion exchange chromatography, IC)分別定量氫離子及酸根離子對手套的滲透質量,並驗證以廣用試紙做為職業安全衛生人員

現場評估之半定量工具之可行性。同時以人工汗液為收集介質,評估其對酸性溶液滲透行為之影響。材料與方法:本實驗之挑戰溶液為硝酸(65%及33%)及硫酸(96%及18%),收集介質為去離子水及人工汗液。測試不同厚度之丁腈橡膠(Ansell Solvex)手套 - A (0.38 mm)、B (0.56 mm)和氯丁橡膠(MAPA)手套 - C (0.60 mm)、D (0.75 mm)。滲透測試以恆溫水浴控制溫度為27C,每組滲透實驗皆進行三重複,每15分鐘採樣一次持續4小時。以pH meter及廣用試紙同步測量挑戰溶液之pH值,偵測氫離子(H+)的破出時間(Breakthrough Time,

BT),以IC定量滲透樣本之酸根離子質量並定義破出時間(BTs),分別計算所有離子的累積滲透質量(Cumulated Permeated Mass, CPM)。以單因子變異數分析(ANOVA)檢定相同材質及型號之手套樣本密度之差異;配對t-test檢定不同量測工具(pH meter vs. 廣用試紙)之差異;Mann-Whitney U test檢定不同收集介質之差異,統計顯著性定義為p-Value  0.05。結果:於4小時之實驗中,所有挑戰溶液對氯丁橡膠手套D (0.75 mm),本研究厚度最厚的手套,均未觀察到破出(BT > 240 分鐘)。以65%硝酸為挑戰溶液,丁腈橡膠手套A平均破

出時間為50 ± 8.66分,B為125 ± 22.91分鐘,氯丁橡膠手套皆未破出;以96%硫酸為挑戰試溶液時,二個材質的表現相反,較厚的氯丁橡膠手套C 的平均破出時間為60 ± 30分鐘,較丁腈橡膠手套(A 110 ± 8.66分鐘,B 175±8.66分鐘)短;低濃度的酸性溶液(18%硫酸及33%硝酸)挑戰丁腈橡膠手套A,本研究厚度最薄的手套,則未偵測到破出。本研究所使用丁腈橡膠手套A、B及氯丁橡膠手套C、D其密度皆具有統計上差異(p  0.05)。以人工汗液或去離子水為收集介質時,發現96%硫酸及65%硝酸對手套A的破出時間無統計上顯著差異(p = 0.7、0.2)。pH meter和

廣用試紙所偵測之破出時間也無統計上顯著差異(p = 0.149)。結論:影響酸性溶液滲透的首要因子是材質及厚度,氫離子濃度則是此二酸性溶液中影響破出時間(BT)的關鍵離子。建議職業安全衛生人員選擇化學防護手套可依循廠商提供之數據,依所使用的酸性溶液選擇適當材質,不可僅以所謂耐酸鹼手套做為通用的防護手套。廣用試紙可做作業現場即時評估手套防護效能的檢點工具。人工汗液不會影響硫酸及硝酸對手套的滲透行為。

空腸彎曲桿菌在雞肉產品加工過程中的交叉污染及抗菌物質對其之抑制效果

Campylobacter spp. 已被公認為近世紀以來最常引起腸胃炎之人畜共通病原菌,屬於食源性病原體。近年來,隨著食品加工之需求的增長,使食品工業操作衛生的重要性大為提高,尤其是加工分切階段、透過與被污染的食品接觸的器材道具,容易發生交叉污染現象。本研究之目的擬以禽肉分切場之作業流程中不同溫度之下所發生交叉污染的機率,及探討乳酸菌片球菌素和香辛料 (大蒜、生薑、洋蔥) 之抗菌效果,以及與螯合劑 (EDTA) 合併使用之組合對雞肉產品之空腸彎曲桿菌的抗菌效果。實驗中所採用四種不同溫度 (10°C、15°C、20°C、25°C) 以及八種不同情境來模擬交叉污染與各種器具 (手套、刀子、砧板

以及解凍水) 之接觸。此外,探討溫度和各種器具材質對空腸彎曲桿菌之轉移率的影響。另外抗菌測試不同濃度香辛料之萃取物 (10%、25%、50%、75% 和100%) 與探討產生 pediocin 之乳酸菌 Pediococcus pentosaceus BCRC 14024 對空腸彎曲桿菌之抑菌效果。結果顯示隨著溫度的上升,空腸彎曲桿菌之轉移率也逐漸上升,實驗中最高之轉移率為在25°C從被污染的雞肉轉移到砧板 (29.40%),而相同使用浸泡過受污染的雞屠體之水源持續浸泡未受污染的雞肉在於25°C 也具有相當高之轉移率 (27.50%)。大蒜在100% 的濃度顯示為最佳的抑制結果、其抑菌環為46

.50±3.5 mm,而生薑與洋蔥在最高濃度的抑制效果分別為25.00±0.0 mm與17.00±1.0 mm。產Pediocin之乳酸菌BCRC 14024最適培養時間為42小時,培養溫度為30°C,培養之起始pH值為5,在此培養條件下所產之pediocin對於Campylobacter jejuni具有較佳之抑菌效果。另外,乳酸菌片球菌素若與EDTA合併使用具對C. jejuni之抑菌能力大於單一使用、其抑菌環直徑為 14±0.8 mm。透過風險計算空腸彎曲桿菌之暴露量為0.178 log CFU/6-kg package,最後計算出消費者在收到本案例公司之產品當下,能被驗出該菌在雞肉產

品中之風險值為 0.00814。本篇研究說明雞肉在加工過程中存在相當程度的交叉污染以及各種抗菌物質的抑菌效果之研究可為雞肉產品在加工過程中的風險管理策略上提供科學依據。
